Rohit scored his ninth ODI ton to take India to an imposing total of 309/3. But a 242-run third wicket partnership between Smith and George Bailey (112) took the match away from the visitors.
Opting to bat first, India were helped by a brilliant 209-run stand between Rohit and Kohli (91), the fifth highest for India against Australia in ODIs.
Australia started their chase on a poor note as they lost openers Aaron Finch (8) and David Warner (5) to debutant pacer Barinder Sran but Smith and Bailey arrested the slide.
They started on a slow not and then gradually increased the run-rate. The stand was finally broken by Ashwin in the 42nd over as Bailey was caught by Bhuvneshwar Kumar. The Indian spinner gave hope as he scalped Glenn Maxwell (6) next.
Maxwell was caught by Shikhar Dhawan as Australia lost their fourth wicket with 37 runs needed in 38 balls. Smith, however, ensured that there was no further hiccup as he took his team to a win and 1-0 lead in the five-match series.
